Se estudió el efecto de regímenes de temperatura y tratamiento pre-germinativos sobre el porcentaje (PG), velocidad (VG) de germinación y actividad enzimática de β-glucosidasa, α-D-galactosidasa, β-1,3-glucanasa y α-amilasa de Crotalaria longirostrata y Lupinus exaltatus. Semillas maduras de ambas especies se recolectaron en la región de los Valles y Libres del Serdán, Puebla (L. exaltatus) y en Macuspana, Tabasco (C. longirostrata). El régimen de temperatura mostró un efecto altamente significativo en el PG y actividad de α-D-galactosidasa en ambas especies, y no se detectó actividad de α-amilasa y β-1,3-glucanasa. En L. exaltatus, el mayor PG y VG se presentó en el régimen de 25 °C/25 °C con 25.92 % y 0.68 semillas día-1; y en C. longirostrata en el régimen de 30 °C/20 °C con 6.11 % y 0.20 semillas día-1. En esta especie, la mayor actividad de α-D-galactosidasa se observó en el régimen 7 días a 5 °C seguido de 20 °C/20 °C (10 547.5 U/mg proteína total); y en β-glucosidasa en el régimen de 20 °C/15 °C (215.6 U/mg de proteína total). En C. longirostrata, la mayor actividad, ambas enzimas, se presentó en el régimen de 20 °C/15 °C; 16 162.3 U/mg proteína total y 539.3 U/mg de proteína total de α-D-galactosidasa y β-glucosidasa, respectivamente. Los tratamientos pre-germinativos con H2SO4 al 98 % por 15 min y H2O a 80 °C por 1 min incrementaron la absorción de agua, PG y VG en L. exaltatus y C. longirostrata, respectivamente. A las 120 h después de siembra, las semillas de L. exaltatus alcanzaron un PG y VG de 30.95 % y 0.07 semillas h-1, respectivamente; las de C. longirostrata de 55.56 % y 0.19 semillas h-1. En ambas especies, la actividad de la α-D-galactosidasa fue siempre mayor en semillas enteras y las de α-amilasa y β-1,3-glucanasa fueron extremadamente bajas en ambas especies (< 0.1 U/mg de proteína total). Después de la aplicación del tratamiento pre-germinativo donde se presentó el mayor PG, también se observó la mayor actividad de α-D-galactosidasa. En C. longirostrata, fue a las 72 h en el tratamiento de H2O 80 °C 1 min (82 624.2 U/mg de proteína total) y en L. exaltatus a las 36 h en el tratamiento H2SO4, 98 % 15 min (9 521.3 U/mg de proteína total). En contraste, no se observó relación entre actividad de β-glucosidasa y PG. La mayor actividad de esta enzima se observó a la hora 0 en C. longirostrata (tratamiento H2SO4, 98 % 15 min con 511.3 U/mg de proteína total), mientras que en L. exaltatus se presentó en el tratamiento de 35 °C/25 °C (172.1 U/mg de proteína total). _______________ CHANGES IN ENZYME ACTIVITY OF GERMINATED AND UNGERMINATED SEEDS OF WILD LEGUMES. ABSTRACT: The effect of temperature regime and pre-germinative treatments were studied on the percentage (GP), speed (GS) of germination and enzyme activity of β-glucosidase, α-D-galactosidase, β-1,3-glucanase and α-amylase in Lupinus exaltatus and Crotalaria longirostrata. Dry seeds of both species were collected in the region of Valles del Serdán and Libres, Puebla (L. exaltatus) and Macuspana, Tabasco (C. longirostrata). The temperature regime showed a highly significant effect on the PG and activity of α-D-galactosidase in both species. No α-amylase y β-1,3-glucanase enzyme activity was detected. In L. exaltatus, the highest GP and GS was observed in the regime of 25 °C/25 °C with 25.92 % and 0.68 seeds days-1 and C. longirostrata in the regime of 30 °C/20 °C with 6.11 % and 0.20 seeds day-1. In this specie, an increase of α-D-galactosidase activity was observed in the regime 7 days at 5 °C followed by 20 °C/20 °C (10 547.5 U/mg total protein); and β-glucosidase in the regime of 20 °C/15 °C (215.6 U/mg total protein). In C. longirostrata, the highest activity, both enzymes, was observed in the regime of 20 °C/15 °C; 16 162.3 U/mg total protein (α-D-galactosidase) and 539.3 U/mg total protein (β-glucosidase). The pre-germinative treatment 15 min with 98 % H2SO4 and H2O at 80 °C for 1 min increased water absorption, GP and SG and enzyme activity. At 120 h after seeding, the L. exaltatus seeds reached 30.95 % of GP and a SG of 0.07 seeds h-1; 55.56 % and 0.19 seeds h-1 for C. longirostrata. In both species, the activity of the α-D-galactosidase was higher in the dry seeds and of α-amylase and β-1,3-glucanase were extremely low (< 0.1 U/mg total protein). After application of the pre-germinative treatment that increased GP significatively, a higher α-D-galactosidase activity was also observed. In C. longirostrata, was 72 h after seeding in H2O 80 ° C 1 min treatment (82 624.2 U/mg total protein) and in L. exaltatus at 36 h in H2SO4 98 % 15 min treatment (9 521.3 U/mg total protein). In contrast, no relationship was observed between β-glucosidase activity and GP in both species. Increased activity of this enzyme was observed at time 0 in C. longirostrata (H2SO4 98 % 15 min, 511.3 U/mg total protein), whereas in L. exaltatus was at the treatment 35 °C/25 °C (172.1 U/mg total protein).
