Cholinergic components have been characterized in mammalian lymphocytes, which could play an important role in the regulation of the immune response. Organophosphate pesticides, being inhibitors of the enzyme acetylcholinesterase, can exert an immunotoxic effect through the alteration of this extra-neuronal cholinergic lymphocyte system, thus altering the physiology of the lymphocytes. In the present work the tilapia nilótica fish (Oreochromis niloticus) was used as a model organism to evaluate the effect of diazinón (organophosphate pesticide) on the mitochondrial membrane potential in lymphocytes. The fish were exposed to sublethal concentrations of diazinon (3.97, 1.95 and 0.97 ppm) for a period of 6 h, later the splenic tissue was extracted and disintegrated. Mononuclear cells were separated by density gradient, DiOC2 (20 nM) was added for 15 min at 28°C and samples were analyzed by flow cytometry. Preliminary results showed that diazinon at concentrations of 0.97, 1.95 and 3.97 ppm causes loss of membrane potential of 19, 30 and 23 % respectively in comparison to the control group.
En linfocitos de mamíferos se han caracterizado componentes colinérgicos, los cuales podrían tener un papel importante en la regulación de la respuesta inmune. Los plaguicidas organofosforados, al ser inhibidores de la enzima acetilcolinesterasa, pueden ejercer un efecto inmunotóxico a través de la alteración de este sistema colinérgico extra-neuronal linfocitario, alterando así la fisiología de los linfocitos. En el presente trabajo se utilizó al pez tilapia nilótica (Oreochromis niloticus), como organismo modelo para evaluar el efecto de diazinón (plaguicida organofosforado) sobre el potencial de membrana mitocondrial en linfocitos. Los peces fueron expuestos a concentraciones subletales de diazinón (3.97, 1.95 y 0.97 ppm) por un periodo de 6 h, posteriormente se extrajo y se disgregó el tejido esplénico. Las células mononucleares se separaron por gradiente de densidad, se adicionó DiOC2 (20 nM) por 15 min a 28°C y las muestras se analizaron por citometría de flujo. Los resultados preliminares, mostraron que diazinón a concentraciones de 0.97, 1.95 y 3.97 ppm causa pérdida del potencial de membrana del 19, 30 y 23 % respectivamente en comparación del grupo control.
