COMPARACION DE LAS PRUEBAS ROSA DE BENGALA Y RIVANOL CON ELISA PARA EL DIAGNOSTICO DE BRUCELOSIS BOVINA

Brucellosis is a zoonotic disease that can have a strong impact on the economy and it represents a public health problem. In Mexico, the official diagnosis is made mandatory by serology, using the card test, rivanol and complement fixation, considering the isolation and characterization of Brucella spp as the "gold standard". In this study serological techniques card test (CT) and rivanol (RIV) viceverse ELISA (Enzyme Linked immunoabsorbent Assay) were compared, for the qualitative and quantitative diagnosis of brucellosis. It were used sera from 50 female adult cattle from the municipal slaughterhouse of which 25 showed negative test to CT and 25 showed positive test results to CT and RIV with different titers. It was determined too the relative sensitivity and specificity of the CT and RIV test with respect to ELISA test. Matching it was determined using kappa analysis. CT showed a sensitivity of 96% and a specificity of 100%. RIV it had a sensitivity of 88%. Matching of kappa test between CT and ELISA it was 0.96; between RIV and ELISA of 0.88, but the significance of X2 test it was P < 0.07 when comparing sensitivity of RIV, suggesting that samples with low titers of RIV could represents vaccine antibodies, which ELISA it considers as a negative samples. ELISA is a complementary test for positive animals that can improve the sensitivity in the diagnosis of Brucella abortus.

La brucelosis es una enfermedad zoonótica que puede impactar fuertemente en el ámbito económico, además de representar un problema de salud pública. En México, el diagnóstico oficial obligatorio se realiza por serología, mediante las pruebas de tarjeta rosa de bengala, rivanol y fijación de complemento, considerándose el aislamiento y caracterización de Brucella spp como la “prueba de oro”. En este estudio se comparó el resultado de las técnicas serológicas de tarjeta (RB) y rivanol (RIV) con la prueba de ELISA (Enzime Linked Immunoabsorbent Assay) cualitativo y cuantitativo para el diagnostico de la brucelosis. Se utilizaron sueros sanguíneos de 50 hembras de bovinos adultos procedentes del rastro municipal de las cuales 25 mostraron resultados negativos a la prueba RB y 25 con resultados positivos a las pruebas RB y RIV con diferentes titulaciones. Se determinó la sensibilidad y la especificidad relativa de las pruebas con respecto a ELISA-I. La concordancia se determino mediante el análisis kappa. RB mostró una sensibilidad de 96% y una especificidad del 100%. RIV tuvo una sensibilidad del 88%. La concondancia (kappa) entre RB y ELISA-I fue de 0.96, entre RIV y ELISA-I de 0.88; sin embargo a la prueba de X2 hay una significancia P < 0.07 al comparar la sensibilidad de RIV, sugiriendo que las muestras con títulos bajos a RIV pudieran representar anticuerpos vacunales que ELISA-I considera muestras negativas. ELISA-I es una prueba complementaria para animales positivos que puede mejorar la sensibilidad en el diagnóstico de Brucella abortus.
