"Two enzymatic treatments were tested (commercial mixtures Viscozyme and Celluclast from Novozymes) for saccharification of the polysaccharides present in cooked untreated and pretreated bagasse of Agave tequilana Weber. The highest concentrations of fermentable sugars were obtained from the enzymatic hydrolysis of untreated bagasse. In batch tests, the enzymatic hydrolysates of untreated bagasse showed the best molar yield (HMY) with Celluclast (1.31 moles H2/mol substrate) and the best volumetric hydrogen production rate (VHPR) with Viscozyme (142.48 ml H2/L-h). The fermentation of stabilizing agents (sorbitol or sucrose) present in the enzyme preparations contributed to bio-hydrogen production. The maximum contribution of these stabilizing agents was 46.9% for the hydrolyzates obtained with Celluclast (sorbitol) and 80% for the hydrolyzates obtained with Viscozyme (sucrose). Finally a completed stirred reactor (CSTR) was fed with enzymatic hydrolyzate (Celluclast) from untreated bagasse. In the starting stage, the hydraulic retention times (HRT) of 6 to 24 hours enabled an adequate selection of acidogenic microorganisms, because no methane was detected in the biogas during the operation time. In the evaluation stage, a HMY of 0.76 and 0.74 moles H2/mol substrate and VHPR of 72.55 and 49.56 ml H2/L-h for HRT of 12 and 24 hours respectively were observed. Comparison of HMY and VHPR obtained in this study with the HMY and VHPR values reported in the literature, for other lignocellulosic hydrolysates (0.38-1.70 mol H2/mol sugars; 4.6-81.4 ml H2/L-h), indicates that the enzymatic hydrolyzate of untreated bagasse is a suitable substrate for hydrogen production. The yields and rates of hydrogen found in this work are similar with those reported in the literature, but are lower than those found for model substrates (1.03-2.79 mol H2/mol glucose; 170-1270 ml H2/L-h)."
