Estrés osmótico durante la germinación, emergencia y desarrollo fenológico del maíz

"Se estudió el efecto del potencial osmótico en los procesos fisiológicos relacionados con la capacidad de germinación, vigor, desarrollo vegetativo y variables asociadas con la actividad fotosintética en una variedad experimental de maíz amarillo. La investigación incluyo cuatro estudios, llevados a cabo en el Laboratorio de Fisiología de semillas del Centro de Capacitación y Desarrollo en Tecnología de Granos y Semillas (CCDTS) y en invernadero, ambos pertenecientes a la Universidad Autónoma Agraria Antonio Narro, en Buenavista Saltillo, Coahuila, México. Estudio preliminar: incluyo un ensayo de germinación de semillas y una caracterización física y análisis bromatológico. Estudio I. Ensayo de germinación y vigor, consistió en la siembra de semillas en papel Anchor humedecido con soluciones salinas ajustadas a diez diferentes potenciales osmóticos (PO) en un rango de -0.125 MPa a -1.250 MPa, usando NaCl y un testigo con agua destilada, posteriormente se colocaron en una cámara de germinación (25 ±1°C) por siete días. El vigor y la capacidad germinación se midieron al cuarto y séptimo día después de la fecha de siembra respectivamente. Estudio II. Índice de Velocidad de Emergencia (IVE) las semillas se sembraron en suelo pesado en un invernadero, al inicio del ensayo se aplicaron siete potenciales osmóticos (-0.25, -0.50, -0.75, -1.0, -1.25, -1.5) y un testigo con agua destilada. La emergencia de plántulas se evalúo por siete días, al medio día y al final la se obtuvo la emergencia total (ET) y el peso de las plántulas (PSP). Estudio III. Se evaluaron los caracteres de la planta y variables fotosintéticas en diferentes etapas fenológicas (en las hojas 5, 8, 12, 16) bajo condiciones de estrés salino en invernadero. Se utilizó un medidor portátil de fotosíntesis (LICOR LI-6400) para determinar la tasa de la asimilación de CO2, CO2 intercelular, conductancia estomática (gs) y la tasa de transpiración (E). Los resultados del estudio preliminar mostraron 100% de germinación de la semilla, 10.57 % de proteína, 5.1% de lípidos, 80.92 % de carbohidratos y 1.95 % de contenido de fibra. Estudio I. Los resultados indican que esta variedad de maíz tiene la habilidad de germinar a un PO por encima de -1.00 MPa (CE= 18.57 dSm-1), sin que presente una reducción drástica en el número de x plántulas normales. Sin embargo a PO más bajos, la germinación y el vigor de semillas, así como la longitud de plúmula y de la raíz, así como el peso seco, se reducen en una forma dramática. Estudio II. Reducciones en el PO causa una disminución en el índice de velocidad de emergencia (IVE), emergencia total (ET) y peso seco de plántulas (PS); a -1.5 MPa, el IVE se redujo de 6.26 a 0.09, la ET descendió de 100 a 2.66 % y el PS de 214.35 a 51.5 mg plántula-1. Estudio III. La tasa neta de fotosíntesis no cambio al reducirse el PO de la solución, sin embargo, la conductancia estomática bajo cuando las plantas se regaron con soluciones salinas de -3 MPa, esta respuesta sugiere que la anatomía de la hoja cambio, conservando cloroplastos para llevar a cabo la asimilación de CO2. El Contenido de clorofila bajo 35% cuando un estrés extremo fue aplicado (-3 MPa) pero la hoja pudo mantener la actividad fotosintética. En general, el potencial osmótico de las soluciones tuvo un efecto negativo en la germinación y vigor, así como en la tasa fotosintética. La reducción en la germinación de las semillas expuestas a condiciones de salinidad es resultado del estrés osmótico o la toxicidad iónica que afectan el metabolismo y la movilización de reservas almacenadas en la semilla. Por otra parte cuando las soluciones salinas son aplicadas en invernadero en plantas con un desarrollo más avanzado, la solución de agua afecto el crecimiento por dos razones: 1) se reduce de la habilidad de la planta para absorber agua y esto conlleva reducciones en la tasa de crecimiento, y 2) Cuando grandes cantidades de sal entran en la planta en el flujo de la transpiración, se va a presentar daño celular que resulta en reducciones en el crecimiento. El daño celular causa una disminución en la cantidad de clorofila. Sin embargo la tasa de asimilación de CO2 se mantiene."

"The effect of the osmotic potential on the physiological processes related to germination capacity, vigor, vegetative development and variables associated to the photosynthetic activity was studied, in a yellow maize experimental variety. The research included four studies, carried out in the Seed Physiology Laboratory of the Training and Seed Technology Development Center and in the greenhouse at the Universidad Autónoma Agraria Antonio Narro, in Buenavista, Saltillo, Coahuila, Mexico. Preliminary Study: Included a seed germination test and a physical and bromatological characterization. Study I. Germination and vigor tests, consisted in planting seeds in germination Anchor paper, wetted with salinity solutions and adjusted to ten different osmotic potentials (OP), ranging from -0.125 to -1.250 MPa using NaCl and a check with distilled water. Thereafter were placed in a growth chamber (25±1°C) for 7 days. Vigor and germination capacity were evaluated 4th and 7th days after the planting date, respectively. Study II. Emergence speed index (ESI). Seeds were planted in a greenhouse in a heavy soil, seven saline solutions treatments (- 0.250, -0.500, 0.750, -1.000, -1.250, -1.500 MPa) and a check (distilled water) were applied once at the beginning of the test. Seedlings emergence was evaluated at midday for seven days, and at the end the total emergence (TE) and the seedlings weight (SWT) were obtained. Study III. The responses of agronomic and photosynthetic variables were evaluated in different phenological stages (at the 5th, 8th, 12th and 16th leaf) under saline stress, in greenhouse conditions. A portable photosynthesis meter (LICOR LI-6400) was used, to determine CO2 assimilation rate, intercellular CO2, stomatal conductance and transpiration rate. Results from the preliminary test showed 100 % seed germination, 10.57 % protein, 5.1 % lipids, 80.92 % carbohydrates and 1.95 % fiber content. Study I. Results indicated that this maize variety has the ability to germinate at a PO above -1.00 MPa (CE = 18.57 dS/m), without having a drastic reduction in the number of normal plants. However at lower OP, seed vigor and germination, as well as root and shoot length and dry weight, decreased in a dramatic way. Study II. Decreases in the OP, caused a reduction in the ESI, TE, and SWT; at 1.5 MPa, the ESI dropped from 6.26 (check) to 0.09. The TE descended from xiii 100 to 2.66 % and the SWT from 214.35 to 51.5 mg/seedling. Study III. The net photosynthesis rate did not change with decreasing the OP, however the stomatal conductance dropped when the plants were watered with -3 MPa saline solutions, this response suggests the leaves anatomy changed, conserving chloroplasts to carry out the CO2 assimilation. The chlorophyll content dropped 35 % when extreme stress was applied (-3 MPa), but it was able to maintain the leaf photosynthetic activity. In general, the OP solutions had a negative effect in the seed vigor and germination, and in the photosynthetic rate. Germination breakdown in seeds exposed to saline conditions is the result of the osmotic stress or the ion toxicity, affecting the seed metabolism and the mobilization of stored reserves. On the other hand, when saline solutions are applied to greenhouse grown plants, the water solution affects growth for two main reasons: 1). It reduces the ability of the plant to take up water, and this leads to reductions in the growth rate, and 2). When enormous amounts of salt enter the plant in the transpiration stream, there will cause cell damage, resulting also in growth reductions. Cell damage causes a reduction in the amount of chlorophyll, however the CO2 assimilation rate is constant."
