Rho-type GTPases are key regulators that control eukaryotic cell polarity, but their role in fungal morphogenesis is only beginning to emerge. In this study, we investigate the role of the CDC-42 - RAC - CDC-24 module in Neurospora crassa. rac and cdc-42 deletion mutants are viable, but generate highly compact colonies with severe morphological defects. Double mutants carrying conditional and loss of function alleles of rac and cdc-42 are lethal, indicating that both GTPases share at least one common essential function. The defects of the GTPase mutants are phenocopied by deletion and conditional alleles of the guanine exchange factor (GEF) cdc-24, and in vitro GDP-GTP exchange assays identify CDC-24 as specific GEF for both CDC-42 and RAC. In vivo confocal microscopy shows that this module is organized as membrane-associated cap that covers the hyphal apex. However, the specific localization patterns of the three proteins are distinct, indicating different functions of RAC and CDC-42 within the hyphal tip. CDC-42 localized as confined apical membrane-associated crescent, while RAC labeled a membrane-associated ring excluding the region labeled by CDC42. The GEF CDC-24 occupied a strategic position, localizing as broad apical membrane-associated crescent and in the apical cytosol excluding the Spitzenkörper. RAC and CDC-42 also display distinct localization patterns during branch initiation and germ tube formation, with CDC-42 accumulating at the plasma membrane before RAC. Together with the distinct cellular defects of rac and cdc-42 mutants, these localizations suggest that CDC-42 is more important for polarity establishment, while the primary function of RAC may be maintaining polarity. In summary, this study identifies CDC-24 as essential regulator for RAC and CDC-42 that have common and distinct functions during polarity establishment and maintenance of cell polarity in N. crassa. © 2011 Araujo-Palomares et al. “We place our hope in the land”: defense of the territory against mining in Ixtacamaxtitlán, Puebla
Veronica Vazquez_Garcia Esteban Martínez Vásquez (2023, [Artículo, Artículo])
Mexico occupies first place in mining exploration in Latin America, a situation that has caused alarm among native peoples because the implementation of extractive projects threatens their vital space. The objective of this paper is to analyze the actions of Corporate Social Responsibility (CSR) of the Ixtaca Project in Ixtacamaxtitlán, Puebla, as well as the strategies of territorial defense undertaken by the communities to stop its expansion. Data was gathered through one survey, one workshop and various interviews conducted in three communities affected by the project. Results discuss three actions of CSR: 1) job offers; 2) investment in community infrastructure; 3) support for social events. Three main defense strategies were identified: 1) collective agreements refusing to sell water to the company; 2) legal litigations in favor of the right to be consulted as native peoples; 3) the strengthening of peasant agriculture thanks to the defense of the ejido, the conservation of irrigation water and the support received from the federal program Sembrando Vida. The paper concludes that, despite the negative impacts of RCS actions, these strategies have proven effective because mining concessions have been cancelled and some families have returned to agriculture.

Whole-genome comparison between reference sequences and oyster Vibrio vulnificus C-genotype strains
CARLOS ABRAHAM GUERRERO RUIZ (2019, [Artículo])
Whole-genome sequences of Vibrio vulnificus clinical genotype (C-genotype) from the CICESE Culture Collection, isolated from oysters, were compared with reference sequences of CMCP6 and YJ016 V. vulnificus C-genotype strains of clinical origin. The RAST web server estimated the whole genome to be ~4.8 Mb in CICESE strain 316 and ~4.7 Mb in CICESE strain 325. No plasmids were detected in the CICESE strains. Based on a phylogenetic tree that was constructed with the whole-genome results, we observed high similarity between the reference sequences and oyster C-genotype isolates and a sharp contrast with environmental genotype (E-genotype) reference sequences, indicating that the differences between the C- and E-genotypes do not necessarily correspond to their isolation origin. The CICESE strains share 3488 genes (63.2%) with the YJ016 strain and 3500 genes (63.9%) with the CMCP6 strain. A total of 237 pathogenicity associated genes were selected from reference clinical strains, where—92 genes were from CMCP6, 126 genes from YJ016, and 19 from MO6-24/ O; the presence or absence of these genes was recorded for the CICESE strains. Of the 92 genes that were selected for CMCP6, 67 were present in both CICESE strains, as were as 86 of the 126 YJ016 genes and 13 of the 19 MO6-24/O genes. The detection of elements that are related to virulence in CICESE strains—such as the RTX gene cluster, vvhA and vvpE, the type IV pili cluster, the XII genomic island, and the viuB genes, suggests that environmental isolates with the C-genotype, have significant potential for infection.

Physiological ecology of Mexican CAM plants: history, progress, and opportunities
Joel David Flores Rivas Oscar Briones Villareal JOSE LUIS ANDRADE (2022, [Artículo])
"In Mexico, plants with crassulacean acid metabolism (CAM) are part of the Mexican culture, have different uses and are even emblematic. Unfortunately, only a small fraction of the Mexican CAM plants has been studied physiologically. For this review, the following questions were considered: What ecophysiological studies have been conducted with CAM species native to Mexico? What ecophysiological processes in Mexican CAM plants are the most studied? What type of ecophysiological studies with CAM plants are still needed? A database of scientific studies on CAM plant species from Mexico was documented, including field and laboratory works for species widely distributed, and those studies made outside Mexico with Mexican species. Physiological processes were grouped as germination, photosynthesis, and water relations. Most studies were done for CAM species of Cactaceae, Bromeliaceae, Asparagaceae and Orchidaceae, andmost ecophysiological studies have been done on germination of cacti. Field and laboratory studies on photosynthesis and water relations were mostly for terrestrial cacti and epiphytic bromeliads. There were few physiological studies with CAM seedlings in Mexico and few studies using stable isotopes of water and carbon of CAM plants in the field. More field and laboratory studies of physiological responses and plasticity of CAM plants to multiple stress factors are required to model plant responses to global climate change. In general, more physiological studies are essential for all CAM species and for species of the genus Clusia, with C3-CAM and CAM members, which can become ecologically important under some climate change scenarios."
