RNA Purity, Real-Time PCR Sensitivity, and Colon Segment Influence mRNA Relative Expression in Murine Dextran Sodium Sulfate Experimental Colitis.

The dextran sodium sulfate (DSS) model of colitis is widely used as a result of its simplicity and reproducibility and because it mimics clinicopathological disease features. Its effectiveness depends on the mouse strain, DSS MW, and brand. Quantitative RT-PCR (qRT-PCR) is highly sensitive for analyzing cytokine mRNA expression. We analyzed an acute model of DSS treatment in Balb/c mice for the onset of colitis using qRT-PCR for the quantification of a mouse cytokine transcript. We compared differences among 1--and 2-step qRT-PCR for transcript quantification, the effect of multiple concentrations of DSS, and the use of 2 reference genes in 3 portions of the colon. A reliable and sensitive 1-step protocol for qRT-PCR was established with a modified double LiCl precipitation for RNA isolation. The variability of 2 reference genes, ?-actin and eukaryotic elongation factor 2, was compared, and expression of IL-6 was analyzed in 3 segments of the colon. The RNA cleaning protocol prevented inhibition of qRT-PCR by DSS, and RNA loss was minimized. No clinical differences among the different DSS concentrations were seen on d 7, but higher concentrations resulted in the appearance of earlier symptoms. Higher efficiency and sensitivity of the 1-step qRT-PCR reaction using eukaryotic elongation factor 2 were obtained and also less variability. Although expression levels of IL-6 were high in the middle and distal colon, the middle section had consistently less variability in values. Thus, this segment is recommended for future studies. These factors influence the statistical significance of data and need to be considered to get accurate and reliable results and to improve comparisons of the published colitis experiments.

"Proteins of the Split ends (Spen) family are characterized by an N-terminal domain, with one or more RNA recognition motifs and a SPOC domain. In Arabidopsis thaliana, the Spen protein FPA is involved in the control of flowering time as a component of an autonomous pathway independent of photoperiod. The A. thaliana genome encodes another gene for a putative Spen protein at the locus At4g12640, herein named AtSpen2. Bioinformatics analysis of the AtSPEN2 SPOC domain revealed low sequence similarity with the FPA SPOC domain, which was markedly lower than that found in other Spen proteins from unrelated plant species. To provide experimental information about the function of AtSpen2, A. thaliana plants were transformed with gene constructs of its promoter region with uidA::gfp reporter genes; the expression was observed in vascular tissues of leaves and roots, as well as in ovules and developing embryos. There was absence of a notable phenotype in knockout and overexpressing lines, suggesting that its function in plants might be specific to certain endogenous or environmental conditions. Our results suggest that the function of Atspen2 diverged from that of fpa due in part to their different transcription expression pattern and divergence of the regulatory SPOC domain."

"A novel bipartite begomovirus, Blechum interveinal chlorosis virus (BleICV), was characterized at the genome level. Comparative analyses revealed that BleICV coat protein (CP) gene promoter is highly divergent from the equivalent region of other begomoviruses (BGVs), with the single exception of Tomato chino La Paz virus (ToChLPV) with which it shares a 23-bp phylogenetic footprint exhibiting dyad symmetry. Systematic examination of the homologous CP promoter segment of 132 New World BGVs revealed the existence of a quasi-palindromic DNA segment displaying a strongly conserved ACTT-(N7)-AAGT core. The spacer sequence between the palindromic motifs is constant in length, but its sequence is highly variable among viral species, presenting a relaxed consensus (TT)GGKCCCY, which is similar to the Conserved Late Element or CLE (GTGGTCCC), a putative TrAP-responsive element. The homologous CP promoter region of Old World BGVs exhibited a distinct organization, with the putative TATA-box overlapping the left half of the ACTT-N7 composite element. Similar CP promoter sequences, dubbed “TATA-associated composite element” or TACE, were found in viruses belonging to different Geminiviridae genera, hence hinting unsuspected evolutionary relationships among those lineages. To get cues about the TACE function, the regulatory function of the CLE was explored in distinct experimental systems. Transgenic tobacco plants harboring a GUS reporter gene driven by a promoter composed by CLE multimers expressed high beta-glucuronidase activity in absence of viral factors, and that expression was increased by begomovirus infection. On the other hand, the TrAP-responsiveness of a truncated CP promoter of Tomato golden mosaic virus (TGMV) was abolished by site-directed mutation of the only CLE present in it, whereas the artificial addition of one CLE to the -125 truncated promoter strongly enhanced the transactivation level in tobacco protoplasts. These results indicate that the CLE is a TrAP-responsive element, hence providing valuable clues to interpret the recurrent association of the CLE with the TACE. On the basis of the aforesaid direct evidences and the insights afforded by the extensive comparative analysis of BleICV CP promoter, we propose that the TACE might be involved in the TrAP-mediated derepression of CP gene in vascular tissues."

Identificación de ovinos pelibuey resistentes a la infección por Haemonchus contortus en función de parámetros inmunitarios.

El objetivo del presente estudio fue analizar los rasgos fenotípicos y la expresión relativa de genes asociados a la respuesta inmune contra la infección por el nematodo hematófago H. contortus en corderos Pelibuey resistentes a la hemoncosis. Se compararon dos modelos de segregación para identificar a los corderos resistentes en función del conteo fecal de huevos (FEC, por sus siglas en inglés) expresado como huevos por gramo de heces (EPG, por sus siglas en inglés). El primer método considerado de referencia fue la  ± 3 EE de EPG y en el segundo método se dividió la muestra usando cuartiles. El primer método fue más sensible para identificar 22 corderas resistentes, 10 de susceptibles y ocho resilientes de un total de 40 animales. En el segundo ensayo experimental fueron segregados nueve corderos con fenotipo de resistencia, seis de susceptibles y 14 resilientes a la infección por H. contortus. En los corderos resistentes se observó una reducción mayor del 90% en el FEC. Posteriormente, se seleccionaron cinco corderos susceptibles y resistentes, respectivamente, para llevar a cabo los estudios de expresión relativa de genes. Las citocinas (interleucinas, IL) IL-2, IL-4, IL-5, IL-6, IL-8, IL-10, IL-13, IFN-γ, TGFβ1 relacionados con la respuesta inmune y el receptor FCεR1A de IgE fueron seleccionados para analizar el nivel de expresión. Los resultados mostraron incremento en el número de transcriptos de RNAm de IL-4, IL-5 y FCεR1A en corderos resistentes con respecto a los susceptibles. Además, se observó correlación negativa entre el gen IL-5 con el FEC. Los resultados obtenidos sugieren la función biológica del gen IL-5 como marcador genético y la importancia biológica de los genes IL-4 y del receptor FCεR1A en ovinos Pelibuey resistentes para regular la infección por el nematodo parásito H. contortus. _______________ IDENTIFICATION OF PELIBUEY RESISTANT SHEEP TO INFECTION BY Haemonchus contortus IN FUNCTION IMMUNE PARAMETERS. ABSTRACT: The aim of the present study was to determine the phenotype traits and relative expression of immune genes against the blood-feeding nematode H. contortus in Pelibuey sheep-breed resistant to parasite. The comparison of two segregation models was used to identify resistant lambs based on the faecal egg count (FEC) indicated as the number of eggs per gram (EPG). The first method  ± 3 EE of EPG was considered as reference and the second method was divided in quartiles. The first method was more sensitive to identify 22-lambs with resistance phenotype traits, 10-lambs with susceptible traits and eight with resilient traits from a total 40-lambs. In the second experimental assay nine-lambs were segregated as resistance, six susceptible and 14 resilient to H. contortus infection. The resilient lambs had 90% of FEC reduction was observed. Then, five resistant and susceptible lambs were selected to performance the relative expression studies. Nine cytokines (Interleukins, IL) IL-2, -4, -5, -6, -8, -10, -13, IFN-γ, TGFβ1 genes related with the immune-response and the FCεR1A receptor of IgE were selected to analyse the expression level. Results showed increased expression of mRNA transcripts of IL-4, IL-5 and FCεR1A in resistant lambs compared with susceptible-lambs with decreased expression. In addition, negative correlation was observed between IL-5 gene and FEC. These results suggest the biological role of IL-5 gene as molecular marker and the importance of IL-4 gene and FCεR1A receptor in resistant Pelibuey-lambs against the main parasitic nematode, H. contortus.

The goals of our study were to determine the possible association of interleukin (IL)-31 with Th17 cytokine profile in serum and to quantify retinoic acid-related orphan receptor C (RORC) mRNA expression in psoriatic arthritis (PsA) patients. This cross-sectional study was conducted in 50 patients with PsA and 30 control subjects (CS) matched by age and gender. The cytokine serum levels were quantified by magnetic bead - based assay using the Bio-Plex MAGPIX system, and RORC mRNA expression was determined by quantitative polymerase chain reaction (qPCR).

Identification of differential expressed transcripts in cervical cancer of Mexican patients

"The aim of this study was to identify the gene expression profile in biopsies of patients with cervical intraepithelial neoplasia (CIN) 1, CIN 2, CIN 3, and microinvasive cancer by suppression subtractive hybridization and Southern blotting. After analyzing 1,800 cDNA clones, we found 198 upregulated genes, 166 downregulated, and no significant change of gene expression in 86 clones (p = 0.005). These results were validated by Northern blot analysis (p = 0.0001) in the identification of 28 overexpressed and 7 downregulated transcripts. We observed a set of genes related to the Notch signaling pathway that may be involved in the transformation of cervical cells and in the development to malignancy. The differentially expressed genes may provide useful information about the molecular mechanisms involved in human cervical carcinoma and as diagnostic markers."

Evaluación de las concentraciones salivales de IL-6 en pacientes con enfermedad periodontal con y sin diabetes mellitus tipo 2

La enfermedad periodontal (EP) es una condición inflamatoria que estimula la secreción de IL-6. Esta citocina ha sido relacionada con la destrucción del periodonto y un estado de inflamación de bajo grado sistémico que favorece el desarrollo de diabetes mellitus tipo 2 (DM2), no obstante, los valores reportados en adultos con EP y DM2 son discordantes. El objetivo del estudio fue determinar las concentraciones salivales de IL-6 en adultos con EP con y sin DM2.

Antecedentes. La enfermedad periodontal (EP) es una condición inflamatoria que estimula la secreción de IL-6. Esta citocina ha sido relacionada con la destrucción del periodonto y un estado de inflamación de bajo grado sistémico que favorece el desarrollo de diabetes mellitus tipo 2 (DM2), no obstante, los valores reportados en adultos con EP y DM2 son discordantes. El objetivo del estudio fue determinar las concentraciones salivales de IL-6 en adultos con EP con y sin DM2. Métodos. Se realizó un estudio transversal descriptivo que incluyó 90 participantes asignados a tres grupos: 30 con DM2 y EP (DM2+EP), 30 sin DM2 con EP (EP) y 30 sin DM2 y sin EP (no DM2-EP). Se llevó a cabo una historia clínica, medición de peso y talla, examen periodontal, evaluaciones bioquímicas (glucosa, triglicéridos, colesterol total, HDL colesterol) y determinación de IL-6 salival. A través de pruebas chi2 y ANOVA o KruskalWallis se compararon las características clínicas y antropométricas entre grupos, y mediante pruebas Mann-Whitney-U-Wilcoxon se compararon los valores de IL-6 entre grupos según éstas características. Se ajustó un modelo de regresión lineal estratificado por edad para contrastar los valores de IL-6 del grupo DM2+EP frente a los grupos EP y no DM2-EP. Resultados. Los participantes del grupo DM2+EP presentaron mayores concentraciones salivales de IL-6 (1.7±4.5 pg/dL) que aquéllos de los grupos EP (0.6±2.1 pg/dL) y no DM2- EP (1.4±1.7 pg/dL). Asimismo, los individuos con obesidad y con DM2+EP presentaron valores más altos de IL-6 (1.2±3.3 pg/dL, rango=0-11.4) que aquéllos no DM2-EP con obesidad (3.3±0.9 pg/dL, rango=0-4.2; p=0.027). Mediante análisis de regresión lineal 44 ajustado por sexo, IMC y HDL-colesterol, los valores de IL-6 fueron mayores en los participantes DM2+EP de 39-60 años de edad que en los individuos EP del mismo rango de edad (β=3.03, IC95%: 0.172, 5.899); al incluir otras covariables, esta relación no fue significativa. Conclusiones. Las concentraciones salivales de IL-6 son mayores en adultos de 39 a 60 años de edad que presentan DM2 y EP que aquéllos que presentan únicamente EP del mismo rango de edad. Estos resultados sugieren un vínculo entre la DM2 y EP con un estado inflamatorio crónico.

La infección por Helicobacter pylori es el principal factor de riesgo para cáncer gástrico; en la respuesta inmune inducida por la bacteria existe un infiltrado de células que secretan citocinas, favoreciendo el desarrollo de la carcinogénesis gástrica. El objetivo fue evaluar la relación de la expresión de IFN-γ e IL-4 con la infección por H. pylori, gastritis crónica y cáncer gástrico. Se incluyeron 100 pacientes con gastritis crónica y 20 con cáncer gástrico; se determinó la expresión de INF-γ e IL-4 por inmunohistoquímica. El 57.5% de los casos fueron positivos a H. pylori, la mayor frecuencia de expresión de IFN-γ se encontró en los ca-sos de cáncer positivos a la bacteria, en el caso de IL-4 se encontró más frecuente en gastritis crónica H. pylori positivo. Se localizó mayor frecuencia de expresión de IFN-γ en comparación con IL-4; en la mayoría de las muestras positivas a H. pylori expresaron las dos citocinas

The Helicobacter pylori infection is the main factor of risk for gastric cancer, in the im-mune response induced for the bacterium there is an infiltration of cells which produce high levels of cytokines, which may be involved in gastric carcinogenesis. The objetive of this investigation is to evaluate the relation of the expression of IFN-γ and IL-4 with the H. pylori infection, chronic gastritis and gastric cancer. 100 patients with chronic gastritis and twenty with gastric cancer were included; the expression of INF-γ and IL-4 was determined by immunohistochemistry. 57.5% of cases were positive for H. pylori, the highest frequency of IFN-γ expression was found in cases of cancer H. pylori-positive, in the case of IL-4 was more frequent in patients H. pylori-positive with chronic gastritis.
