Coronin is a component of the endocytic collar of hyphae of neurospora crassa and is necessary for normal growth and morphogenesis

Coronin plays a major role in the organization and dynamics of actin in yeast. To investigate the role of coronin in a filamentous fungus (Neurospora crassa), we examined its subcellular localization using fluorescent proteins and the phenotypic consequences of coronin gene (crn-1) deletion in hyphal morphogenesis, Spitzenkörper behavior and endocytosis. Coronin-GFP was localized in patches, forming a subapical collar near the hyphal apex; significantly, it was absent from the apex. The subapical patches of coronin colocalized with fimbrin, Arp2/3 complex, and actin, altogether comprising the endocytic collar. Deletion of crn-1 resulted in reduced hyphal growth rates, distorted hyphal morphology, uneven wall thickness, and delayed establishment of polarity during germination; it also affected growth directionality and increased branching. The Spitzenkörper of Δcrn-1 mutant was unstable; it appeared and disappeared intermittently giving rise to periods of hyphoid-like and isotropic growth respectively. Uptake of FM4-64 in Δcrn-1 mutant indicated a partial disruption in endocytosis. These observations underscore coronin as an important component of F-actin remodeling in N. crassa. Although coronin is not essential in this fungus, its deletion influenced negatively the operation of the actin cytoskeleton involved in the orderly deployment of the apical growth apparatus, thus preventing normal hyphal growth and morphogenesis. © 2012 Echauri-Espinosa et al.

Effect of antimicrobial nanocomposites on Vibrio cholerae lifestyles: Pellicle biofilm, planktonic and surface-attached biofilm

Vibrio cholerae is an important human pathogen causing intestinal disease with a high incidence in developing countries. V. cholerae can switch between planktonic and biofilm lifestyles. Biofilm formation is determinant for transmission, virulence and antibiotic resistance. Due to the enhanced antibiotic resistance observed by bacterial pathogens, antimicrobial nanomaterials have been used to combat infections by stopping bacterial growth and preventing biofilm formation. In this study, the effect of the nanocomposites zeolite-embedded silver (Ag), copper (Cu), or zinc (Zn) nanoparticles (NPs) was evaluated in V. cholerae planktonic cells, and in two biofilm states: pellicle biofilm (PB), formed between air-liquid interphase, and surface-attached biofilm (SB), formed at solid-liquid interfaces. Each nanocomposite type had a distinctive antimicrobial effect altering each V. cholerae lifestyles differently. The ZEO-AgNPs nanocomposite inhibited PB formation at 4 μg/ml, and prevented SB formation and eliminated planktonic cells at 8 μg/ml. In contrast, the nanocomposites ZEO-CuNPs and ZEO-ZnNPs affect V. cholerae viability but did not completely avoid bacterial growth. At transcriptional level, depending on the nanoparticles and biofilm type, nanocomposites modified the relative expression of the vpsL, rbmA and bap1, genes involved in biofilm formation. Furthermore, the relative abundance of the outer membrane proteins OmpT, OmpU, OmpA and OmpW also differs among treatments in PB and SB. This work provides a basis for further study of the nanomaterials effect at structural, genetic and proteomic levels to understand the response mechanisms of V. cholerae against metallic nanoparticles. © 2019 Meza-Villezcas et al. Screening of xylose utilizing and high lipid producing yeast strains as a potential candidate for industrial application

"Background: Sustainable production of oil for food, feed, fuels and other lipid-based chemicals is essential to meet the demand of the increasing human population. Consequently, novel and sustainable resources such as lignocel- lulosic hydrolysates and processes involving these must be explored. In this paper we screened for naturally-occurring xylose utilizing oleaginous yeasts as cell factories for lipid production, since pentose sugar catabolism plays a major role in efcient utilization of lignocellulosic feedstocks. Glycerol utilization, which is also benefcial in yeast-based oil production as glycerol is a common by-product of biodiesel production, was investigated as well. Natural yeast isolates were studied for lipid accumulation on a variety of substrates, and the highest lipid accumulating strains were further investigated in shake fask cultivations and fermenter studies on xylose and hydrolysate. Results: By collecting leaves from exotic plants in greenhouses and selective cultivation on xylose, a high frequency of oleaginous yeasts was obtained (>40%). Diferent cultivation conditions lead to diferences in fatty acid contents and compositions, resulting in a set of strains that can be used to select candidate production strains for diferent purposes. In this study, the most prominent strains were identifed as Pseudozyma hubeiensis BOT-O and Rhodosporidium toruloides BOT-A2. The fatty acid levels per cell dry weight after cultivation in a nitrogen limited medium with either glucose, xylose or glycerol as carbon source, respectively, were 46.8, 43.2 and 38.9% for P. hubeiensis BOT-O, and 40.4, 27.3 and 42.1% for BOT-A2. Furthermore, BOT-A2 accumulated 45.1% fatty acids per cell dry weight in a natural plant hydrolysate, and P. hubeiensis BOT-O showed simultaneous glucose and xylose consumption with similar growth rates on both carbon sources. The fatty acid analysis demonstrated both long chain and poly-unsaturated fatty acids, depending on strain and medium. Conclusions: We found various natural yeast isolates with high lipid production capabilities and the ability to grow not only on glucose, but also xylose, glycerol and natural plant hydrolysate. R. toruloides BOT-A2 and P. hubeiensis BOT-O specifcally showed great potential as production strains with high levels of storage lipids and comparable growth to that on glucose on various other substrates, especially compared to currently used lipid production strains..."

El concepto, la circunstancia de la enseñanza que aquí abordaremos, tiene el propósito de restituir ciertas situaciones que corresponden al propio ejercicio de la instrucción y a aquellas otras que hablan del acontecer, por ello el trabajo. Tiene las variables tanto de la experiencia real como de las otras sobre las que se han escrito, una de ellas, al final, plantea una situación paradójica que se refiere a la del maestro ignorante que, básicamente, tiene como propósito mostrar que el conocimiento es algo que está en el alumno y que el docente debe limitarse a tratar de que aflore y se exprese de la mejor manera posible. Nos habla de la independencia y de las características que puede tener una educación rigurosa y rígida, en contraposición con una más flexible. Lo mismo sucede con el pensamiento desde nuestra América, de la educación dependiente a una educación libertaria.

The concept, the circumstance of the teaching that we will address here has the purpose of restoring certain situations that correspond to the exercise itself as those others that speak of the happening for it the work. It has the variables of real experience as of the others about which they have been written, one of them, in the end, poses a paradoxical situation that refers to that of the ignorant teacher that, basically, has the purpose of showing that knowledge is something that is in the student and that the teacher must limit himself to trying to bring it out and express itself in the best possible way. It tells us about independence and the characteristics that a rigorous and rigid education can have with a more flexible one and the same happens with the thought from our America, from dependent education to a libertarian education.

Los frutos secos comúnmente conocidos como nueces, son alimentos importantes en la dieta cotidiana debido a su rico sabor y su valioso aporte nutricional. Sin embargo, al querer degustar este tipo de alimentos nos enfrentamos a la problemática de romper la cáscara, la cual se considera un material duro. De lo anterior surge la pregunta: ¿Por qué las cáscaras de nueces son tan duras? En este artículo se discutirán las características (morfológicas, microestructurales y composición) que tienen estos alimentos y que dan como resultado que exhiban un alto grado de rigidez.

El objetivo principal de este estudio fue identificar la razón de la disminución de la capacidad de conducción de la línea 1 y 2 y proponer alternativas para recuperar dicha capacidad. Los tramos de TSI-TP y TP-TASJ operan por debajo de su capacidad de conducción y tienen una parte operando como canal. El “cuello de botella” está en el tramo TO5-TSI en el cual se puede presentar alguna de las siguientes situaciones: aire atrapado; obstrucciones físicas en la tubería; alta rugosidad de las paredes de la conducción.
