The Pacific harbor seal gut microbiota in Mexico: Its relationship with diet and functional inferences

Diet is a primary driver of the composition of gut microbiota and is considered one of the main routes of microbial colonization. Prey identification is fundamental for correlating the diet with the presence of particular microbial groups. The present study examined how diet influenced the composition and function of the gut microbiota of the Pacific harbor seal (Phoca vitulina richardii) in order to better understand the role of prey consumption in shaping its microbiota. This species is a good indicator of the quality of the local environment due to both its foraging and haul-out site fidelity. DNA was extracted from 20 fecal samples collected from five harbor seal colonies located in Baja California, Mexico. The V4 region of 16S rRNA gene was amplified and sequenced using the Illumina technology. Results showed that the gut microbiota of the harbor seals was dominated by the phyla Firmicutes (37%), Bacteroidetes (26%) and Fusobacteria (26%) and revealed significant differences in its composition among the colonies. Funtional analysis using the PICRUSt software suggests a high number of pathways involved in the basal metabolism, such as those for carbohydrates (22%) and amino acids (20%), and those related to the degradation of persistent environmental pollutants. In addition, a DNA metabarcoding analysis of the same samples, via the amplification and sequencing of the mtRNA 16S and rRNA 18S genes, was used to identify the prey consumed by harbor seals revealing the consumption of prey with mainly demersal habits. Functional redundancy in the seal gut microbiota was observed, irrespective of diet or location. Our results indicate that the frequency of occurrence of specific prey in the harbor seal diet plays an important role in shaping the composition of the gut microbiota of harbor seals by influencing the relative abundance of specific groups of gut microorganisms. A significant relationship was found among diet, gut microbiota composition and OTUs assigned to a particular metabolic pathway. © 2019 Pacheco-Sandoval et al. High quality diet improves lipid metabolic profile and breeding performance in the blue-footed booby, a long-lived seabird

Understanding the role of diet in the physiological condition of adults during reproduction and hence its effect on reproductive performance is fundamental to understand reproductive strategies in long-lived animals. In birds, little is known about the influence of the quality of food consumed at the beginning of the reproductive period and its short-term effects on reproductive performance. To assess the role of diet in the physiological condition of female blue-footed booby, Sula nebouxii (BFBO), during reproduction we evaluated whether individual differences in diet (assessed by using δ13C and δ15N values of whole blood from female birds and muscle tissue of the principal prey species) prior to egg laying and during incubation influenced their lipid metabolic profile (measured as triglyceride levels and C:N ratio) and their reproductive performance (defined by laying date, clutch size and hatching success). Females with higher δ15N values in their blood during the courtship and incubation periods had a higher lipid metabolic profile, earlier laying date, greater clutch size (2–3 eggs) and higher hatching success. Females that laid earlier and more eggs (2–3 eggs) consumed more Pacific anchoveta (Cetengraulis mysticetus) and Pacific thread herring (Opisthonema libertate) than did other females. These two prey species also had high amounts of lipids (C:N ratio) and caloric content (Kcal/g fresh weight). The quality of food consumed by females at the beginning of reproduction affected their physiological condition, as well as their short-term reproductive performance. Our work emphasizes the importance of determining the influence of food quality during reproduction to understand the reproductive decisions and consequences in long-lived animals. © 2018 González-Medina et al. Phylogenetic relationships of Pseudo-nitzschia subpacifica (Bacillariophyceae) from the Mexican Pacific, and its production of domoic acid in culture

Pseudo-nitzschia is a cosmopolitan genus, some species of which can produce domoic acid (DA), a neurotoxin responsible for the Amnesic Shellfish Poisoning (ASP). In this study, we identified P. subpacifica for the first time in Todos Santos Bay and Manzanillo Bay, in the Mexican Pacific using SEM and molecular methods. Isolates from Todos Santos Bay were cultivated under conditions of phosphate sufficiency and deficiency at 16°C and 22°C to evaluate the production of DA. This toxin was detected in the particulate (DAp) and dissolved (DAd) fractions of the cultures during the exponential and stationary phases of growth of the cultures. The highest DA concentration was detected during the exponential phase grown in cells maintained in P-deficient medium at 16°C (1.14 ± 0.08 ng mL-1 DAd and 4.71 ± 1.11 × 10−5 ng cell-1 of DAp). In P-sufficient cultures DA was higher in cells maintained at 16°C (0.25 ± 0.05 ng mL-1 DAd and 9.41 ± 1.23 × 10−7 ng cell-1 of DAp) than in cells cultured at 22°C. Therefore, we confirm that P. subpacifica can produce DA, especially under P-limited conditions that could be associated with extraordinary oceanographic events such as the 2013–2016 "Blob" in the northeastern Pacific Ocean. This event altered local oceanographic conditions and possibly generated the presence of potential harmful species in areas with economic importance on the Mexican Pacific coast. © 2020 Quijano-Scheggia et al. Protein retention assessment of four levels of poultry by-product substitution of fishmeal in rainbow trout (Oncorhynchus mykiss) diets using stable isotopes of nitrogen (δ15N) as natural tracers

This is second part from an experiment where the nitrogen retention of poultry by-product meal (PBM) compared to fishmeal (FM) was evaluated using traditional indices. Here a quantitative method using stable isotope ratios of nitrogen (δ15N values) as natural tracers of nitrogen incorporation into fish biomass is assessed. Juvenile rainbow trout (Oncorhynchus mykiss) were fed for 80 days on isotopically distinct diets in which 0, 33, 66 and 100% of FM as main protein source was replaced by PBM. The diets were isonitrogenous, isolipidic and similar in gross energy content. Fish in all treatments reached isotopic equilibrium by the end of the experiment. Two-source isotope mixing models that incorporated the isotopic composition of FM and PBM as well as that of formulated feeds, empirically derived trophic discrimination factors and the isotopic composition of fish that had reached isotopic equilibrium to the diets were used to obtain a quantitative estimate of the retention of each source of nitrogen. Fish fed the diets with 33 and 66% replacement of FM by PBM retained poultry by-product meal roughly in proportion to its level of inclusion in the diets, whereas no differences were detected in the protein efficiency ratio. Coupled with the similar biomass gain of fishes fed the different diets, our results support the inclusion of PBM as replacement for fishmeal in aquaculture feeds. A re-feeding experiment in which all fish were fed a diet of 100% FM for 28 days indicated isotopic turnover occurred very fast, providing further support for the potential of isotopic ratios as tracers of the retention of specific protein sources into fish tissues. La cuarentena sanitaria de los últimos dos años (COVID-19) obligó a “rediseñar”, “digitalizar” o “virtualizar” nuestros cursos y estilos de enseñanza en tiempo récord para “adaptarlas” al “formato virtual a distancia” (online). La profusión de comillas en las líneas anteriores tiene la intención de señalar el carácter incierto o erróneo que ciertos términos-fetiche adquirieron en el proceso. Es importante desmitificar esas figuras de la expresión que se elevaron casi al nivel de pseudo-epistemes (presuntamente novedosas) para la enseñanza del Diseño. Ese tema lo abordo con mayor amplitud en una nueva investigación, recientemente registrada en la UAM. En este texto esbozo (en forma algo lúdica) pero seria y breve a la vez, la conveniencia de tal desmitificación y avanzo en el delineamiento de un modelo de la alternancia y convergencia de recursos pedagógico-didácticos síncronos y asíncronos, mediante la tecnología-red digital, como un elemento estratégico para la educación futura en Diseño.

The health quarantine of the last 2 years (COVID-19) forced us to “redesign”, “digitize” or “virtualize” our courses and teaching styles in record time to “adapt” them to the “virtual format” “at a distance” (online). The profusion of quotation marks in the previous lines is intended to indicate the uncertain or erroneous character that certain fetish-terms acquired in the process. It is important to demystify those figures of expression that have risen almost to the level of pseudo-epistemes (presumably novel) for teaching design. I address this topic more fully in a new investigation, recently registered at the UAM. In this text I outline (in a somewhat playful way) but serious and brief at the same time, the convenience of such “demystification” and I advance in the outline of a model of alternation and convergence of synchronous and asynchronous pedagogical-didactic resources, through technology. -digital network, as a strategic element for future design education.

Realidad virtual en fenómenos del espacio interestelar

El proyecto es un prototipo que se distribuye en varias etapas y sigue un enfoque de diseño de videojuegos para crear una experiencia de realidad virtual (también conocida como VR Virtual Reality) organizada y coherente. Requiere una fusión de conceptos multidisciplinarios, incluyendo ingeniería, diseño tridimensional y programación, lo que demanda a los profesionales involucrados tener sólidos conocimientos y habilidades creativas. El diseño de videojuegos se valora como un proceso que exige dedicación y pasión, aunque también se destaca la importancia de establecer reglas y géneros para orientar el desarrollo del juego. En este proyecto, el juego se clasifica como una aventura en primera persona centrada en la exploración del espacio interestelar, donde el jugador debe seguir reglas preestablecidas para alcanzar objetivos. La ludología, como estudio académico de los juegos, resalta la constante esencia de jugar, aprender y socializar en los juegos. La construcción del mundo del juego se basa en un Game Design Document que describe la visión, género y objetivos del juego. "Space Travel" se centra en la exploración espacial y cuenta con una nave espacial minimalista y un exoplaneta rocoso y gélido. Las mecánicas del juego se centran en la recolección de objetos y su activación, con reglas que guían la interacción del jugador con el entorno. Este proyecto busca ofrecer una experiencia de VR inmersiva y atractiva, centrada en el usuario. El enfoque en los principios de diseño de videojuegos y la planificación a través del Game Design Document demuestran un compromiso con la creación de una experiencia gratificante y significativa. La atención a la inmersión y las mecánicas de juego respaldan la afirmación de que es un desarrollo centrado en la experiencia del usuario.

The ongoing project is a prototype in various stages, following a game design approach to create an organized and coherent virtual reality experience. It requires a fusion of multidisciplinary concepts, including engineering, three-dimensional design, and programming, demanding that involved professionals possess strong knowledge and creative skills. Game design is valued as a process that demands dedication and passion, while also emphasizing the importance of establishing rules and genres to guide game development. In this project, the game is classified as a first-person adventure focused on interstellar space exploration, where the player must adhere to preset rules to achieve objectives. Ludology, as an academic study of games, highlights the constant essence of playing, learning, and socializing within games. The construction of the game world is based on a Game Design Document describing the vision, genre, and objectives of the game. "Space Travel" focuses on space exploration, featuring a minimalist spaceship and an icy, rocky exoplanet. Game mechanics revolve around object collection and activation, with rules guiding the player's interaction with the environment. This project aims to deliver an immersive and engaging virtual reality experience centered on the user. The focus on game design principles and planning through the Game Design Document demonstrates a commitment to creating a rewarding and meaningful experience. Attention to immersion and game mechanics supports the assertion that it's a user experience focused development.

Desarrollo de un entorno virtual para operar de forma segura el proceso de desorción utilizando el método HAZOP

En este trabajo se describe la metodología del desarrollo de un entorno virtual para el monitoreo de la seguridad industrial de un proceso de desorción a escala laboratorio. La metodología HAZOP fue utilizada para la identificación de los riesgos en el proceso de desorción, analizando de manera particular la alimentación del vapor. Los programas de cómputo Blender y Unity fueron utilizados para el desarrollo del entorno virtual y Python y C# para la programación de scripts. Con la interfaz gráfica desarrollada, el usuario pudo establecer las condiciones de operación del proceso de desorción para predecir las situaciones de riesgo y tomar decisiones previo a la operación física del equipo.

This work describes the methodology for the development of a virtual environment to industrial safety monitoring of a desorption process on a laboratory scale. The HAZOP methodology was used to identify risks in the desorption process, specifically the steam feed. The computer programs Blender and Unity were used for the development of the virtual environment and Python and C# for script programming. With the developed graphical interface, the user was able to set the operating conditions of the desorption process to predict risk situations and make decisions before the physical operation of the equipment.
