Enhancement of antibiotics antimicrobial activity due to the silver nanoparticles impact on the cell membrane

The ability of microorganisms to generate resistance outcompetes with the generation of new and efficient antibiotics; therefore, it is critical to develop novel antibiotic agents and treatments to control bacterial infections. An alternative to this worldwide problem is the use of nanomaterials with antimicrobial properties. Silver nanoparticles (AgNPs) have been extensively studied due to their antimicrobial effect in different organisms. In this work, the synergistic antimicrobial effect of AgNPs and conventional antibiotics was assessed in Gram-positive and Gram-negative bacteria. AgNPs minimal inhibitory concentration was 10–12 μg mL-1 in all bacterial strains tested, regardless of their different susceptibility against antibiotics. Interestingly, a synergistic antimicrobial effect was observed when combining AgNPs and kanamycin according to the fractional inhibitory concentration index, FICI: <0.5), an additive effect by combining AgNPs and chloramphenicol (FICI: 0.5 to 1), whereas no effect was found with AgNPs and β-lactam antibiotics combinations. Flow cytometry and TEM analysis showed that sublethal concentrations of AgNPs (6–7 μg mL-1) altered the bacterial membrane potential and caused ultrastructural damage, increasing the cell membrane permeability. No chemical interactions between AgNPs and antibiotics were detected. We propose an experimental supported mechanism of action by which combinatorial effect of antimicrobials drives synergy depending on their specific target, facilitated by membrane alterations generated by AgNPs. Our results provide a deeper understanding about the synergistic mechanism of AgNPs and antibiotics, aiming to combat antimicrobial infections efficiently, especially those by multi-drug resistant microorganisms, in order to mitigate the current crisis due to antibiotic resistance. © 2019 Vazquez-Muñoz et al. Wastewater-containing dyes are an environmental problem. The prime source of dye pollutants is the textile industry, such as paper manufacturing, food processing, leather, pigments, etc. Dye removal from wastewater using nanotechnology has received attention in recent decades thanks to efficient nanomaterials improving traditional technologies. In recent years, multiple research reports on carbon nanotubes for dye removal and photocatalytic dye degradation provided substantial insight into the comprehension of nanotechnology and remediation. This work presents a review and bibliometric analysis of carbon nanotubes for dye removal and photocatalytic dye degradation, which have an environmental impact today. The bibliometric study showed that the current research tendency on carbon nanotubes applied in dye removal and photocatalysis is still growing. According to research, this work observed that carbon nanotubes for dye removal exhibit high removal and efficient photocatalysis activity, indicating the functionality of nanotechnology for environmental remediation. The analysis of the parameters involved in the removal studies, such as temperature and pH, showed adsorption behavior. The photodegradation of methylene blue demonstrated the photocatalytic activity of carbon nanotubes attributed to the sp2 lattice of graphitic configuration.

Multiplication of native endomycorrhizae isolated from arid soils on organic substrates in wheat plants (Triticum aestivum)

"Purpose Organic residues of coffee pulp, sugarcane bagasse and mature bovine manure are a source of organic matter and nutrients for the multiplication of endomycorrhizae consortia. Therefore, the purpose of this research is to multiply the AMFs in such substrates to decrease soil and water pollution. Method A pot experiment under greenhouse conditions was conducted in order to evaluate the influence of agricultural residues (C2-GEC, C3-PAR, C12-PRO, C14-ZAR) with different genera of endomycorrhizae isolated from semi-arid soils, 75 days after the crop was established. Agronomic characteristics and mineral content of N, K, Ca, Mg, and Fe in root and shoot were evaluated in wheat (Triticum aestivum). Results Multiplication of endomycorrhizae was influenced by the residue type. Greater production of spores was ob-served in the coffee pulp, followed by the sugarcane bagasse, where a higher colonization was obtained in combination of C2-GEC and C3-PAR consortia. This consortia combination also was one of those that have increased the content of N, K, Ca, Mg, and Fe in roots and shoots of wheat.Conclusion Combination of native endomycorrhiza substrates and consortia provides an alternative tool that benefits the physiology and nutrition of the plant to be used in sustainable agricultural production systems."

Alternative cropping and feeding options to enhance sustainability of mixed crop-livestock farms in Bangladesh

We investigated alternative cropping and feeding options for large (>10 cows), medium (5–10 cows) and small (≤4 cows) mixed crop – livestock farm types, to enhance economic and environmental performance in Jhenaidha and Meherpur districts – locations with increasing dairy production – in south western Bangladesh. Following focus group discussions with farmers on constraints and opportunities, we collected baseline data from one representative farm from each farm size class per district (six in total) to parameterize the whole-farm model FarmDESIGN. The six modelled farms were subjected to Pareto-based multi-objective (differential evolution algorithm) optimization to generate alternative dairy farm and fodder configurations. The objectives were to maximize farm profit, soil organic matter balance, and feed self-reliance, in addition to minimizing feed costs and soil nitrogen losses as indicators of sustainability. The cropped areas of the six baseline farms ranged from 0.6 to 4.0 ha and milk production per cow was between 1,640 and 3,560 kg year−1. Feed self-reliance was low (17%–57%) and soil N losses were high (74–342 kg ha−1 year−1). Subsequent trade-off analysis showed that increasing profit and soil organic matter balance was associated with higher risks of N losses. However, we found opportunities to improve economic and environmental performance simultaneously. Feed self-reliance could be increased by intensifying cropping and substituting fallow periods with appropriate fodder crops. For the farm type with the largest opportunity space and room to manoeuvre, we identified four strategies. Three strategies could be economically and environmentally benign, showing different opportunities for farm development with locally available resources.

Metabolites Profile of Extracts and Fractions of Erythroxylum mexicanum Kunth by UHPLC-QTOF-MS/MS and its Antibacterial, Cytotoxic and Nitric Oxide Inhibitory Activities

The present study shows the untargeted metabolite profiling and in vitro antibacterial, cytotoxic, and nitric oxide (NO) inhibitory activities of the methanolic leaves extract (MLE) and methanolic stem extract (MSE) of Erythroxylum mexicanum, as well as the fractions from MSE. Using ultra-high performance liquid chromatography/quadrupole time-of-flight tandem mass spectrometry (UHPLC-QTOF-MS/MS), a total of 70 metabolites were identified; mainly alkaloids in the MLE, while the MSE showed a high abundance of diterpenoids. The MSE fractions exhibited differential activity against Gram-positive bacteria. Notably, the hexane fraction (HSF) against Streptococcus pyogenes ATCC 19615 (MIC=62.5 µg/mL) exhibited a bactericidal effect. The MSE fractions exhibited cytotoxicity against all cancer cell lines tested, with selectivity towards them compared to a noncancerous cell line. Particularly, the HSF and chloroform fraction (CSF) showed the highest cytotoxicity against prostate cancer (PC-3) cells, with IC50 values of 19.9 and 18.1 µg/mL and selectivity indexes of 3.8 and 4.2, respectively. Both the HSF and ethyl acetate (EASF) fractions of the MSE inhibited NO production in RAW 264.7 macrophages, with NO production percentages of 50.0% and 51.7%, respectively, at a concentration of 30 µg/mL. These results indicated that E. mexicanum can be a source of antibacterial, cytotoxic, and anti-inflammatory metabolites.

“La presente tesis explora el diseño, descripción, análisis y hallazgos de la investigación respecto a los procesos de normalización y desnormalización del envenenamiento en San Pedro Barrientos (SPB), una problemática profunda que tiene a los cánceres por asbesto como uno de los puntos más crudos y brutales. En la breve presentación doy a conocer la estructura de la tesis, y algunas reflexiones iniciales sobre mi implicación en el trabajo de campo y respecto a la antropología encarnada. La primera parte de la tesis está compuesta por dos capítulos; en el primer capítulo desarrollo el contexto de producción de la investigación y la relevancia que tuvo la convergencia de cinco grandes procesos organizativos para establecer las condiciones de posibilidad del Proyecto de Investigación-Incidencia en SPB y con ello mi aproximación al tema. Reconozco así el modo en que estas genealogías influyeron en el desarrollo, el diseño metodológico de la investigación, expongo el planteamiento del problema, el desarrollo, el diseño metodológico de la investigación y la delimitación de actores en el cual se corresponde con la propuesta teórica en tanto relacional, ecológica, con un énfasis en la observación y participación. En el segundo capítulo formulo el encuadre teórico-conceptual de la investigación el cual, partiendo del enfoque relacional de la antropología médica crítica (AMC), busca describir, analizar y establecer relaciones entre las prácticas de prevención, reparación y atención con bastante centralidad la ecología política del problema de investigación y lo que denominaré como la ecología política de la enfermedad y las prácticas y representaciones del proceso de salud-enfermedad-atención-prevención-reparación (p-s-e-a-p-r) el cual está nutrido de los prolíficos estudios desde la ecología política en salud, una subdisciplina de la ecología política con una larga trayectoria articulando procesos de salud enfermedad con elementos ecológico-políticos”.

"En este artículo se estudia el doble o doppelgänger como tema central en La invención de Morel, de Adolfo Bioy Casares, a partir de los conceptos de doble objetivo y subjetivo (Jourde y Tortonese, 1996). El descubrimiento de la máquina de Morel incita un cambio de dobles en la novela: de uno objetivo a uno subjetivo. La identidad del narrador-fugitivo y su convivencia con las otras imágenes son elementos fundamentales para entender el papel del doble y su modificación en el relato. A pesar de haber sido mencionado por la crítica, ningún artículo había estudiado el doble como tema central de la novela, por lo cual esta investigación presenta una perspectiva innovadora. Microbiological Analysis of the Air in a Popular Fish Processing and Marketing Area

"Fish are marketed as a food and consumed worldwide. During the production of food, contamination by microorganisms is possible through the air, soil, water, surfaces, food handlers, etc. The air does not have a natural microbial composition, but it is a vehicle for the transmission of microorganisms of economic and health interest because they are associated with food spoilage and human diseases. The objective of this study was the microbiological analysis of the air in an area popular for the processing and marketing of fish products in the city of Tepic Nayarit. Using the passive or sedimentation method to collect microorganisms present in the air, the proportion of aerobic mesophile bacteria, coliform bacteria, fungi and yeast was determined at different locations in the fish processing and marketing area for four weeks. The results indicated that the aerobic mesophiles had the highest counts among all the microbial groups analyzed at the twelve different sampling points during the four weeks of the study; their numbers ranged from 2.44 to 2.95 log CFU/m3/h, followed by molds with counts from 1.44 to 2.75 log CFU/m3/h, yeasts with counts from 0.7 to 2.01 log CFU/m3/h and coliforms with counts that ranged from 0.7 to 1.68 log CFU/m3/h. We determined the proportion of the viable microbiological population present in the air at the different sampling points of the study area; several of these sampling points presented values above those recommended by various agencies around the world. Knowledge of the biological hazards transported through the air is important to establish and reduce the risk to the health of occupants and the contamination pathways of processed and marketed fishery products that may be associated with spoilage and foodborne diseases."

Innovative experimental plant to improve coffee processing

In Mexico, coffee production refers to a process with traditional processing practices that combine rustic and semi-mechanized activities, where quality is not controlled, placing it in the category of conventional coffee. In recent years worldwide, the demand for high-quality coffee has increased and in order to offer this model, it is necessary to standardize the processing and transformation processes that increase the quality and value of the coffee. In order to exploit the different capacities in our country, it is necessary to design an innovative experimental plant for the processing of the primary coffee beneficiary, which is capable of adapting to the processes known by the producers, through the instruments and machinery the plant will provide The producer has control over the variables that influence taking advantage of the organoleptic properties of the coffee bean prior to harvest. The plant allows experimentation and obtaining controlled results according to the primary processing method to be applied. The development of the experimental plant has three iterative design cycles; 1) concept design, in which the various processes and methods investigated are evaluated to seek their homogenization with the help of instruments for monitoring and control, 2) prototype design, which is carried out with the help of 2D platforms and 3D modeling, that allow visualizing the location of the different equipment, as well as the flows of the different primary processing processes that the producer wishes to apply, and 3) innovation design, in which the products and services allow the value chain to be improved through processes instruments, data control and new techniques. For the development and maintenance of the experimental plant, it is necessary to link the entities that make up the quadruple helix; companies, government, society and public centers and institutes of higher education. In order to promote teamwork, collaboration and the exchange of ideas to strengthen, sustain and enrich the innovation ecosystem created through the experimental plant.
