Gene expression in primary hemocyte culture of the Pacific white shrimp Penaeus vannamei infected with different white spot syndrome virus (WSSV) strains

"Five previously analyzed white spot syndrome virus (WSSV) strains from northwest Mexico,differing in their genome architecture as well as in virulence, were selected (high virulence JP and LG strains;moderate virulence GVE and DIV strains; and low virulence LC10 strain) to evaluate pathogenesis response in vitro. Expression of phagocytosis-activating protein PAP, manganese superoxide dismutase MnSOD and peroxiredoxin PRX, and two genes of immediate-early expression (IE1 and WSSV304) were measured by qPCR in a primary hemocyte cell culture from Penaeus vannamei at 1, 3, 6, 12, and 24 h post-infection (hpi). PAP expression was significantly higher at 1 and 3 hpi, and JP and LC10 strains induced the highest expression. The response of MnSOD was high at 1 hpi, and a significant increase in PRX expression was detected at 3 hpi, probably due to the occurrence of an oxidative burst; expression levels of MnSOD and PRX were significantly higher at 1 and 3 hpi, respectively, induced by the LG strain (high virulence), suggesting an acute response. In general, expression of most immune-related - genes decreased after the initial hours of infection. Expression levels of IE1 and WSSV304 were exceptionally high at 1 hpi in almost all five WSSV analyzed strains,confirming their efficient mechanism for replication and viral fitness. The results of this study do not show an accurate link between the genome size and WSSV virulence of the strains, albeit the strain with the smallest genome showed the highest virulence. All strains induced an early immune response in heterogeneous ways."

Propuesta de mejora para la integración del equipo de salud en el código azul en áreas críticas adulto

La presente investigación se realizará en un hospital de segundo nivel de atención de la ciudad de Cuernavaca, el código azul es un sistema de alarma que existe en la mayoría de los hospitales, está conformado por un equipo multidisciplinario de salud que se encuentra capacitado y es convocado de manera inmediata con funciones previamente asignadas para poder intervenir de forma oportuna, eficaz y con calidad a pacientes en paro cardiopulmonar. Este trabajo de investigación propone la integración del equipo de salud en código azul.

Se menciona como fue la historia de la formación de los grupos de reanimación cardiopulmonar y la organización del código azul; fue el primer modelo de estos sistemas y ha demostrado un impacto positivo en la morbimortalidad de los pacientes, la conexión de la teórica Patricia Benner de principiante a experto, se describe como se vive un código azul en el área hospitalaria, la necesidad de estructurar tarjetas que identifique que rol tiene cada uno de los integrantes del código azul, el personal de salud tiene que tener capacidad de reconocer la emergencia del evento, realizando oportunamente las intervenciones del código azul. Metodología adecuada para esta investigación: se decidió un estudio de tipo cuantitativo, longitudinal, observacional, cuasiexperimental y de intervención educativa, será dirigido a todo profesional de salud; área médica, enfermería, camilleros, residentes, pasantes y estudiantes de ambas profesiones de los diferentes turnos que laboran en las áreas críticas del hospital. Se pretende aplicar dos instrumentos de evaluación diagnóstica y de habilidades previo a la capacitación curso- taller con la finalidad de fortalecer los conocimientos y habilidades y posteriormente aplicar los mismos instrumentos de evaluación final. Se implementará un pizarrón en cada servicio, donde describa el rol de la actividad que tendrá cada integrante que conforme el equipo de código azul, así mismo se proporcionará el manual de procedimientos de código azul, guía de práctica clínica impresa que se ubicaran en un lugar estratégico para su consulta y gafetes que describa la función que le corresponde a cada uno de los miembros.

This research will be carried out in a second level care hospital in the city of Cuernavaca. The code blue is an alarm system that exists in most hospitals; it is made up of a multidisciplinary health team that is trained and immediately summoned with previously assigned functions in order to intervene in a timely, effective and quality manner in patients in cardiopulmonary arrest. This research work proposes the integration of the health team in code blue.

It mentions the history of the formation of cardiopulmonary resuscitation groups and the organization of the code blue; it was the first model of these systems and has shown a positive impact on patient morbidity and mortality, the connection of the theorist Patricia Benner from beginner to expert, it describes how a code blue is experienced in the hospital area, the need to structure cards that identify the role of each of the members of the code blue, the health personnel must have the ability to recognize the emergency event, performing timely interventions of the code blue. Appropriate methodology for this research: a quantitative, longitudinal, observational, quasi-experimental, quasi-experimental and educational intervention study was decided, and it will be directed to all health professionals; medical area, nursing, orderlies, residents, interns and students of both professions of the different shifts that work in the critical areas of the hospital. It is intended to apply two diagnostic and skills evaluation instruments prior to the training course-workshop in order to strengthen knowledge and skills and then apply the same instruments for the final evaluation. A blackboard will be implemented in each service, describing the role of the activity that each member of the code blue team will have, as well as the code blue procedure manual, printed clinical practice guide that will be placed in a strategic place for consultation and badges that describe the function that corresponds to each of the members.

Association between nonalcoholic fatty liver disease and severe male reproductive organ impairment (Germinal Epithelial Loss): Study on a mouse model and on human patients

Metabolic syndrome (MS) has been associated with testicular damage. Nonalcoholic fatty liver disease (NAFLD) is a multisystemic disease that affects different organs, but its effect on the testes is unknown. A study analyzing germ cell involvement on BALB/c mice was carried out. A parallel comparative study was conducted that investigated alterations in the germinal epithelium of male humans that died from an unrelated acute event. The complete medical histories and histologic samples of the thoracic aorta, liver tissue, and testicular tissue from the deceased subjects were collected. The degree of germinal epithelial loss (DGEL) was evaluated and the clinical and histologic data were compared between individuals with and without NAFLD. The only metabolic or morphologic variable that caused a significant difference in the DGEL, in both the animal model and humans, was the presence of liver steatosis. The percentage of steatosis was also correlated with the percentage of the DGEL. In humans, steatosis (greater than 20%) increased the risk 12-fold for presenting with a severe DGEL (OR: 12.5; 95% CI [1.2, 128.9]; p = .03). There was no association with age above 50 years or MS components. Steatosis grade was also correlated with atherosclerosis grade. NAFLD was a strongly associated factor implicated in severe DGEL, as well as the testis was identified as a probable target organ for damage caused by the disease. his finding could result in the search for new approach strategies in the management of men with fertility problems. Further studies are required to confirm these results.

Estas interacciones pueden actuar sobre la base de ejercer un efecto tóxico, interrumpiendo funciones importantes para el desarrollo celular, como el metabolismo del ácido desoxirribonucleico (ADN) y las proteínas del ciclo celular. De Novo assembly and annotation of the Pacific calico scallop (Argopecten ventricosus) transcriptome for immune-related gene discovery

"Invertebrates' immune defense mechanisms play a critical role in pathogen recognition and elimination. De novo assembly and annotation of the Argopecten ventricosus transcriptome were performed for the immune-related gene identification. Scallops (height: 4.4 cm) were challenged with inactivated Vibrio parahaemolyticus IPNGS16. The RNA from different tissues was pooled for a single cDNA library construction sequenced by NextSeq 500 platform 2×75 paired‐end chemistry. Before de novo assembling with Trinity, reads were analyzed with FastQC, Trimmomatic, and Prinseq. Assembled sequences were analyzed by CD-HIT-EST and TransDecoder. The corresponding annotation was performed against NCBI-nr, RefSeq protein, and KAAS (KEGG) databases. The Trinity assembly yielded 107,516 contigs. TransDecoder yielded 25,285 sequences as CDSs of which, 16,123 were annotated against the NCBI-nr protein, most of them scored with Crassostrea gigas data. Gene ontology mapped sequences (15,262) were classified in molecular functions (~13,000), cellular components (~11,000), and biological processes (~13,000). The KAAS analysis showed biological categories for metabolism (13%), cellular processes (12%), genetic information processing (10%), organismal systems (19%), environmental information (13%), and human diseases (33 %). Within the organismal systems, 467 immune-related genes (KO) were identified. Sixty-four immune-related genes were annotated/blasted against the NCBI-nr and RefSeq protein databases. An RT-qPCR was performed to analyze the expression level of immune-related genes obtained in the transcriptome analysis in scallops (height 4.5 cm) treated with probiotic bacilli added to culture water. Bacilli significantly increased the expression of the HSP70 and PGRP genes. The gene transcripts analysis of A. ventricosus will better understand its immune response against pathogens in culture systems."

Este trabajo de tesis parte de la creciente preocupación global sobre la Epidemia

Metabólica, enfocándose en su prevalencia, impacto económico y las perspectivas

futuras en México. Se centra en la interconexión entre enfermedades metabólicas,

factores de riesgo compartidos y la respuesta a intervenciones comunes,

destacando la obesidad, diabetes y síndrome metabólico como ejes centrales.

Adicionalmente, examina la Medicina Ancestral y la estandarización en la

producción de remedios herbolarios como estrategias potenciales para mitigar esta

crisis.

El trabajo revisa los antecedentes de la Medicina Tradicional según la Organización

Mundial de la Salud (OMS), su uso en condiciones metabólicas, y su integración en

sistemas de salud. Se presta especial atención al uso de plantas medicinales en

México y a nivel global, resaltando la transferencia intercultural de conocimientos

herbolarios en la era de la globalización.

En el marco general, se establecen los objetivos del estudio y se detalla la

caracterización de productos herbales específicos para el tratamiento del síndrome

metabólico, se centra en el uso sinérgico de: Syzigium cumini, Coix lacryma-jobi, y

Mossanona depressa. Se analizan sus propiedades farmacológicas y usos en el

tratamiento de la diabetes y el síndrome metabólico.

La sección de formalización comercial aborda la constitución de la sociedad, los

registros ante el Instituto Mexicano de Propiedad Intelectual (IMPI) y la Comisión

Federal para la Protección contra Riesgos a la Salud (COFEPRIS), la determinación

de patentabilidad, y las estrategias de protección de propiedad intelectual.

Finalmente, el estudio concluye con recomendaciones para la mejora regulatoria en

este campo y detalla la tramitología necesaria para la implementación de estas

estrategias. La investigación subraya la importancia de integrar la medicina

tradicional en los sistemas de salud modernos para combatir la epidemia

metabólica, especialmente en contextos como el de México.

Chronic toxicity of shrimp feed added with silver nanoparticles (Argovit-4®) in Litopenaeus vannamei and immune response to white spot syndrome virus infection

"In recent years, the application of silver nanoparticles (AgNPs) as antibacterial compounds has been widely used in human and veterinary medicine. In this work, we investigated the effects of AgNPs (Argovit-4 R ) as feed additives (feed-AgNPs) on shrimp (Litopenaeus vannamei) using three different methods: 1) chronic toxicity after 28 days of feeding, 2) Effects against white spot syndrome virus (WSSV) challenged by oral route, and 3) transcriptional responses of immune-related genes (PAP, ProPO, CTL-3, Crustin, PEN3, and PEN4) following WSSV infection. The results showed that the feed-AgNPs did not interfere with the growth and survival of shrimp. Also, mild lesions in the hepatopancreas were recorded, proportional to the frequency of the feed-AgNP supply. Challenge test versus WSSV showed that feeding every 7 days with feed-AgNPs reduced mortality, reaching a survival rate of 53%, compared to the survival rates observed in groups fed every 4 days, daily and control groups of feed-AgNPs for the 30%, 10%, and 7% groups, respectively. Feed-AgNPs negatively regulated the expression of PAP, ProPO, and Crustin genes after 28 days of treatment and altered the transcriptional responses of PAP, ProPO, CTL-3, and Crustin after WSSV exposure. The results showed that weekly feeding-AgNPs could partially prevent WSSV infection in shrimp culture. However, whether or not transcriptional responses against pathogens are advantageous remains to be elucidated."
